Description
Bioconjugation reactions are of major interest in biomedical sciences and enable chemists to modify biomolecules in order to give them new functions or properties. This course will cover bioconjugation and biomolecule labeling strategies that enable the exploration of complex biological systems. Practical sessions will illustrate these concepts through examples of bioconjugation of osidic and nucleotide platforms.
Hourly volumes*:
CM: 9 H
Field: 11 a.m.
Objectives
The objective of this course unit is to provide students with theoretical and practical knowledge in the field of bioconjugation reactions and their implementation techniques.
Teaching hours
- Functionalization/Bioconjugation - CMLecture9 a.m.
Mandatory prerequisites
Organic chemistry, M1 level
Knowledge assessment
Final written exam lasting 1 hour 30 minutes
Authorized documents: no
Non-graphing calculator permitted: yes
Internet access: no
Practical work assessed in the form of a group report
Syllabus
Classes: 9 a.m.
- Functional target: Modification of sugars, polysaccharides, and glycoconjugates—modification of nucleic acids and oligonucleotides.
- Chemical modifications of biomolecules, reactivity of the main functional groups (amine, thiol, carboxylate, azide, etc.) of biomolecules. Bioorthogonal ligation reactions.
- Bioconjugation reagents: Homobifunctional, heterobifunctional, and trifunctional links; self-sacrificing probes and tags.
Field: 11 a.m.
The practical sessions (2 x 5.5 hours) carried out in groups will aim to illustrate the concepts detailed in the lectures. The first session will be dedicated to the synthesis of a small functional molecule that will be introduced into a target biomolecule (polysaccharide or oligonucleotide) in a second session. The new compounds synthesized during these practical sessions will also be tested to evaluate their biological activity. Students will therefore be directly involved in an existing research project.
