Description

This course is designed to deepen in small groups the fundamental molecular and cellular processes seen in the BMC2 and BMC3 courses by approaching them through more concrete notions. The lessons will be based on real data (experimental results, scientific articles) to simply explain the main scientific approaches and to learn how to analyze and interpret the results (Example 1: show an interaction in cellulo by expression of tagged proteins in cell lines followed by immunoprecipitation and western-blot. Example 2: principle of immunofluorescence, intracellular distribution of an antigen. Example 3: transcription and translation in vitro and study of interaction by GST-pull down). The practical exercises will illustrate some of these basic approaches: cell culture, construction of expression vectors, transfection, immunolabeling, fluorescence microscopy.

Objectives

- Knowledge:

- knowledge and understanding of the fundamental concepts of cell biology and molecular biology

- understand the principles of basic techniques used in cell biology and molecular biology

- Know-how :

- know how to analyze a figure in a scientific article.

- Know how to analyze and interpret the results of a short/simple scientific article and be able to make a quick synthesis (level 1 of article analysis).

- know how to maintain a cell line in culture

- know how to perform a plasmid transfection on a cell line.

- know how to perform an imunocytochemical / fluorescent labelling.

- Know the basics of using an epifluorescence upright microscope.

- Know how to be :

- work independently and in groups

- Write a report of practical work respecting the rules of presentation and the time allowed

- respect the hygiene and safety rules of a laboratory and the good laboratory practices related to the culture of cell lines

- This UE allows a deepening of the fundamental concepts and techniques of Cellular and Molecular Biology and is essential to follow the L3 Molecular and Cellular Biology course.

-This conceptual and technical deepening course can be interesting in particular for the L3 Biochemistry and Microbiology courses.
